Simply add the required class to the thumbnail
Make sure the href of the thumb points to the actual full image and not the "image page"
I did similar in this example.
i have made some changes to this module:
- title & summary in the picture box
- help info in the picture box
- change in positioning and size of the picture box to work better with big pictures
- new icon with info on mouseover
- new Lightbox library v2.03.3 modified by me to have keyboard navigation in Chrome & Firefox (because in original Esc/arrows keys not working correctly for both browsers)
after instalation the module, first configurate this file: gallery2/modules/lightbox/js/lightbox.js
and set the path to you icon(gif/png) files

There is no mystery to lytebox(s) at this point.
